| Description: | Chronic non-communicable diseases currently represent a major global public health chal-lenge due to their significant contribution to mortality and morbidity. In view of their conti-nuous increase and the limitations of conventional treatments, the exploration of new bioac-tive molecules of natural origin has emerged as a promising approach for the prevention and management of these diseases. This study evaluated the biological activities of the aqueous and hydroethanolic extracts of Senna alexandrina Mill. (Fabaceae), a medicinal plant widely used in traditional medicine for the treatment of digestive disorders. The results showed a higher extraction yield for the hy-droethanolic extract (13.60%) compared with the aqueous extract (9.15%), a difference attri-buted to the extraction solvent and method. Phytochemical screening revealed the presence of various secondary metabolites, including alkaloids, phenolic compounds, flavonoids, sapo-nins, anthraquinones, and quinones, which was further confirmed by thin-layer chromato-graphy. Quantitative analysis demonstrated that the hydroethanolic extract was richer in phe-nolic and flavonoid compounds (127.22 ± 2.18 μg GAE/mg and 33.73 ± 0.18 μg QE/mg, res-pectively).Biological assays revealed significant activities for both extracts. The antioxidant activity (DPPH assay) showed that the hydroethanolic extract exhibited stronger antioxidant activity (IC₅₀ = 10.49 μg/mL) than the aqueous extract (IC₅₀ = 62.05 μg/mL). However, ascorbic acid, used as the positive control, exhibited superior antioxidant activity with an IC₅₀ value of 8.57 μg/mL.The anti-inflammatory assay also confirmed inhibition of protein denaturation, with IC₅₀ va-lues of 526 μg/mL for the hydroethanolic extract and 666 μg/mL for the aqueous extract. So-dium diclofenac, used as the positive control, exhibited higher anti-inflammatory activity with an IC₅₀ value of 443 μg/mL.Both extracts exhibited moderate antidiabetic activity through inhibition of α-amylase and α-glucosidase enzymes, with IC₅₀ values of 507.5 μg/mL for the hydroethanolic extract and 485.7 μg/mL for the aqueous extract, and maximum inhibition rates of 36.84% and 34.97%, respectively. In comparison, acarbose, used as the positive control, showed stronger inhibitory activity, with an IC₅₀ value of 204.04 μg/mL and a maximum inhibition rate of 48.49%.Finally, the thrombolytic assay demonstrated clot lysis rates of 49.1% for the hydroethanolic extract and 40.1% for the aqueous extract at a concentration of 1000 μg/mL. In comparison, heparin, used as the positive control, exhibited a clot lysis rate of 86%.The anticoagulant assay revealed a prolongation of coagulation time, reaching 637 s for the hydroethanolic extract and 434 s for the aqueous extract. In comparison, heparin, used as the positive control, produced a coagulation time of 840 s. In light of these findings, it can be concluded that Senna alexandrina Mill. possesses promi-sing biological properties, mainly attributed to its richness in phenolic and flavonoid com-pounds. These results support its use as a natural source of bioactive compounds and highlight its potential for the future development of therapeutic and preventive products. |
