| Description: | elicobacter pylori, a bacterium implicated in numerous gastric pathologies constitutes a major public health concern. The increasing emergence of strains resistant to conventional antibiotics highlights the need to develop new therapeutic strategies. In this context, peptide deformylase, an enzyme essential for bacterial protein maturation and absent in mammals, represents a particularly promising therapeutic target. The present work aims to identify new potential inhibitors of H. pylori peptide deformylase through a virtual screening approach based on molecular docking using Surflex-Dock software, combined with the evaluation of pharmacokinetic and toxicological properties via the SwissADME and ProTox-II 3.0 platforms. The reliability of the docking protocol was validated by RMSD calculation, indicating a success rate of 74.26%, as well as by visual analysis of the docking poses and the study of the linear correlation between experimental activities and predicted affinity scores (r = 0.715).Among the inhibitors studied, actinonin showed the best binding affinity, with a score of 7.06 M⁻¹, and was selected as the reference molecule for screening 192 structural analogs retrieved from the PubChem database. Compound CID_9879394 emerged as the most promising candidate, with a higher affinity score of 8.42 M⁻¹. Analysis of molecular interactions revealed a binding mode similar to that of actinonin, involving several key active-site residues, namely Gln51, Ile45, Gly46, Leu97, and Glu139. Evaluation of ADME-Tox properties revealed a favorable pharmacokinetic and toxicological profile for this compound, characterized by good oral bioavailability, compliance with Lipinski's rule, no predicted inhibition of the major cytochrome P450 isoforms, and low potential toxicity. The results obtained suggest that compound CID_9879394 represents a promising candidate for the development of new anti-H. pylori agents. However, further complementary in vitro and in vivo experimental studies remain necessary to confirm its biological activity and assess its real therapeutic potential. |
